My wife wants to take a two hour road trip (each direction, total 4hrs) the day after my vasectomy. Is this advised so close to vasectomy?

As long as she is driving you will most likely be ok. Most men the day after are feeling fine although perhaps a little sore. You should not be planning on a lot of activity such as lifting.

As I say often to questions like this is, "the company policy is strictly off your feet the first twenty four hours and then take it easy and off your feet for the most part for an additional forty eight hours." Having said that, if you are doing well the next morning after a vasectomy, riding in a car should not be an issue. It will not affect the vasectomy success rate, but it may predispose you to more swelling, bruising or pain. If you do go and do well, we call that- "the art of getting away with it."
